The pp is right about Brandwine hotels. Not a lot of great options. Philadelphia is a totally different without kids. I met my NYC sister there for a getaway weekend. So many trendy restaurants, shopping, leisurely cups of coffee. Adult drinks at a bar without the nagging tug of responsibility.

Depends which season you go. When we went in the summer, we went tubing and also did yoga in a vineyard. We stayed at the Inn at Montchanin which has a nice restaurant and spa. The restaurant at longwood Gardens is also lovely.
