My son, who loves to paint anything, would absolutely want to paint his nails if he saw me painting mine (which I never do) but my husband would totally flip the hell out. Everyone and their gender needs, ugh.
Reminds me of all the controversy over the JCrew ad a few months back.

There's a little girl at daycare who will be 3 next month and her nails are always painted. I think it's adorable. Not her fingers, only her toes. I think the toes are less likely to go in the mouth at that age.

Just visited a preschool and saw a few of the 4 year old girls with nailpolish, made me wonder when others have started painting their toddler's nails. My two year old is infatuated with my painted nails and toes (well, when I have a manicure). Does anyone paint their toddler's nails as a fun activity? I think she'd LOVE it, but my gut is telling me this is way too young... she could eat off the polish. That said, if you do it, what kind of polish do you use? Anything non toxic?
Toddler, yes. Tween, not so much.

Sorry, but nail polish on any toddler: boy or girl is not a good idea.
Especially when it's not your biological child.
Need to respect what the parents have decided on defining gender roles and support. Seems like you're simply trying to subvert her parenting skills and philosophy by deliberating provoking his mother.
Tread carefully, as a "step parent" of a young child. You have many years ahead of all of you. Don't use the kid as a battle ground.
