Block Island -- perhaps Block Island Inn. Bit more than 6 hours though.
I'll second the NY Finger Lakes recommendation, especially Skaneateles Lake (could stay at Sherwood Inn in village (on the water) but to get real lake experience you should go to VRBO.com and rent a house on the water, but one that's near the village). Drive time is @6 hours going up route 81 - a pretty drive too w/o stress of 95.
Also more than 6 hours -- someplace like the beaches around Charleston, SC, like Seabrook and Kiawah. Drive is @8.5 hours but a pretty drive once you get past the trafficy part of 95 south of DC.

Wilmington is a lovely town. There's a film studio/sound stage there you can take a tour of, they have filmed a lot of movies there.
( you can also check out all the spots in town where they filmed Dawson's Creek)
Wrightsville beach is very cute, you can also check out Kure beach and Carolina beach.
I used to live in Wilmington and miss it greatly. Great beach town. There is fun downtown with a battleship that you can tour. I recommend the serpentarium. I don't like snake but I find this place awesome. There is an aquarium near Carolina Beach. You can rent kayaks or go out on a boat.

Mystic is really small.
It's like a street and a half. There are some bed and breakfasts in the town. There is a hilton close by.
Mystic Pizza is actually pretty good!
There aren't a ton of NY/CT beach towns outside of LBI and the Hamptons. We did a drive down from Cape Cod and stopped at a lot of them. Many are suffering economically.
Cape May has a lot of hotels, it depends on availability.
Is Rhode Island too far? Newport is like 7 hrs

Lavallette, ocean beach, ortley, bay head, pt pleasant. All in nj. Vacancies at area hotels, much quieter since Sandy but everything is rebuilt. Beautiful beaches, old fashioned small beach towns.
