look at flyertalk.com. there are lots of threads about these kinds of selections.
i have not stayed there since it was a marriott, but there is an autograph hotel (the manuna kea I think) in Hawaii that is supposed to be amazing. Also, hard to use points, but we have found a lot of value in several of the vacation club properties. the marriott ones have been VERY nice (although they are not 'luxurious' in a 4 star accommodation way-- with spas etc. They have been great for what they are, and when you stay on a 'hotel' basis there is daily housekeeping.

The Key Biscayne Ritz is a little tired but great for pool and on the beach. You will probably need to drive to dinner.

Ritz Cayman Islands - fantastic spring break. actual room nothing special
Hotel 101 Reykjavik - hotel is nothing amazing but it's probably the nicest hotel in Reykjavik and we really enjoyed Iceland. we stayed here on points and used it as our central point.
W Miami
W Times Square was fun

I think I just like Arizona but I had a good time at the Westin Scottsdale (with kids), Phoenician, JW Marriott and W Scottsdale.

Marriott is really lacking in Miami. I'll check out the W there.

PP here. I forgot about our recent stay at St Regis Bal Harbour. I would highly recommend. Service was fantastic. We got upgraded to a huge suite that overlooked the pool with ocean view.

Vegas is generally cheap for what you get --so probably not a good use of points. Loved the Cosmopolitan though and a ton of nice dining options and next door to the Bellagio.
