I've had issues with depression and anxiety myself and when I am feeling bad I don't find anything funny. Literally. Anyone trying to be funny around me just pisses me off because I'm just trying to get through the day without going off the deep end. Is your wife receiving adequate treatment (meds and therapy?). That would be where I would start. Poor woman - sounds like she is close to a break down.

I am wondering about your jokes/humor. Sarcasm is not funny to a lot of people. It can be grating, passive aggreduce and obnoxious. I would avoid that kind of humor. They say sarcasm is the lowest form of wit. Rent a funny movie and watch it together. Can she laugh at that? I am a somewhat serious person and I cannot stand sarcasm. But I enjoy joking around, late night television and hilarious movies. This year movies that made me laugh out loud include The Interview and Trainwreck.

My guess is you're an unsupportive asshole. That kills a sense of humor.
