This is the type of situation where I encourage people to keep a list of dates, times, witnesses, exact quotes, etc. Oh, and whether or not you're overweight is completely irrelevant.
I would not let it go, first time or not. I think that's horrible advice to tell someone getting picked on at a work place, to let that be her freebie. If I called you, you being HR, would you tell me to let it go and let that be her one screw up?

A while ago, a mother had asked if she thought I'd be interested in babysitting for their family. When the parent came in she said within earshot of me (we were all in the classroom) "no, I wouldn't ask her. She needs a babysitter for herself".
Last week, someone made a comment to me along the lines of "well aren't you special" (I got to leave early that night because all my kids left already) to which my boss said "oh yeah, she's special, but not the kind you're thinking".
Another teacher in my class has noticed this as well and always comments to me that this person seems to be picking solely on me, and that she wouldn't dare do it to someone else.
