I would buy a rental property - small 1 -2 bedroom apartment in a desirable location either locally or at a beach. put 20% down with the money and then rent it out.

The businesses that I want to start cost more than $50K to start. But there are some services that I want that wouldn't cost that much to start, and might fit into the description you gave of a SAHM going back to work.
1) Mildly sick child care: child care for kids who have things like pinkeye...where they are still contagious and can't go to daycare, but aren't terribly sick. Advertise at daycare centers.
2) Services waiting: DH and I had a plumbing problem and we had a choice to either pay the overtime weekend rate or take a day off work to be there during the week. I'd love a service that could meet the plumbers or similar during the week day tasks.

A medical staffing agency...probably mid level providers like NP's or PA's. Many NP jobs are 5 days a week, but the NP's used to be nurses...and nurses work 3 days a week typically. The result is NP's who don't use their degree because of scheduling issues. Such a waste! Many NP's I know would love to work for a staffing agency and just fill in at different offices as needed. Part time NP jobs are super rare.

This has been done but it closed earlier in 2011. It was called the At Play Cafe.

Agree with this post and another that suggested a good indoor play area. Those would be my go-to ideas.
To build on the PP I'm quoting:
-serve healthy, kid friendly foods.
-serve good coffee.
-reasonable prices.

this is pretty brilliant - i often wish whole foods had a drive through option just for this reason...

PP here: I'm thinking akin to Seattle's first coffee kiosks. You would have to offer a very limited but well conceived menu that appealed to the masses of health conscience moms and persnickety kids.

Gluten free cafe (possibly with some vegetarian/vegan and nut free/soy free/egg free options as well). And maybe with a bakery as a part of it.
Because the ability to eat out is nice. But eating out can be tough (and often is a gamble with those who don't really understand).
50K probably is not enough for that. But its something that my sister and I have halfway joked about for a while.
