No, it would not: The Constitution doesn't say anything about the mayor (or about local government in D.C., since the Home Rule Charter wasn't adopted until nearly 200 years after the Constitution was ratified).

It would take a constitutional amendment to give the District of Columbia House and Senate seats. But, it would not take a constitutional amendment to carve out a new state out of everything except the National Mall and adjacent areas.

In particular, it didn't take a constitutional amendment to give a portion of DC back to Virginia, nor did it take constitutional amendments to add states. The constitution does not set a specific size or location for DC.

No. Renaming the mayor would be a statutory change. It wouldn’t change the powers of the person, though, just the name.
