Round Up doesn't linger in the soil. Once the plants are dead you're fine to reseed.

Thick layers of wet newspaper/cardboard kill the grass/weeds and then decompose. Works well for garden areas.

You can buy the landscape fabric at home depot that will kill everything. Also purchase the sod pins so that you can keep the plastic secured over the grass.

If you want to kill everything in that area, why not just buy some Glyphosate concentrate and spray it all down? It'll cost you like $20 for a quart bottle which will easily kill a whole yard's worth of grass and anything else that photosynthesizes.

Look in garden stores for weed-blocking plastic. Or you can just lay down several thicknesses of newspaper or cardboard, weigh it down and wet it. Whatever grows underneath will eventually die off.
