No actually the speed doesn't matter. The person who runs 1 mile around a track will burn 100 calories. The person who walk a mile around a track will burn 100 calories. The difference is that the runner will do it much faster.
Metabolism? Of course it's going to be give or take some depending on age, weight, etc. And yes, walking straight up a hill for a mile will burn more than a flat course. But 100 calories is a good estimate.
