Greenbrier is great. They used to have a great kids club/camp. it's also greatly improved since Jim Justice bought it.
i recently went to the Bedford Springs Resort and it was beautiful. Kind of a mini-Greenbrier. A bit less to do, but a much more manageable weekend trip. Two hours vs. four hours to the Greenbrier.

You can also take the train there, I believe.

The Greenbriar the nicest resort of its type In the east. It's not cheap but in life you often get what you pay for, and the Greenbriar is no exception to that rule. It's not that bad of a drive.

It's too much time in the car for a weekend for my DCs. I've been looking at nemacolin. Closer and cheaper.

Any thoughts on whether this would be a good weekend trip with a 3 year old and 5 year old?
