"I'm not sure" means that you think you know the answer, but are unsure of it.  You know what the possibilities are, but are not sure which of them is correct.

"I don't know" means you don't know the answer, period.

If someone asks you where John Doe lives and you haven't the faintest idea whether it is Kansas or Mumbai, you should say "I don't know" - not "I'm not sure."  Because the latter means you might know the answer, you are just unsure of it. [/quote]
